(MRVP Broth) Buffered Glucose Broth – HX-MEDIA – Hexa Biotech
(MRVP Broth) Buffered Glucose Broth Use :- Recommended fir the Methyl Red and Voges-Proskauer tests in differentiation of the coli-aerogenes group.
Expected Performance when stored at optimum conditions and within the expiry date. No liability accepted for accidents in handling or use.
Directions :- Dissolve 17.00 Grams in 1000 ml distilled water. Boil to dissolve broth the medium completely and distribute aseptically. Sterilize by autoclaving at 15 lbs pressure (121 ‘C) for 15 min, cool it to 42-45 ‘C and inoculate test sample aseptically.
Disposal :- To avoid the contamination or propagation of any hazardous microbes the used, unusable or modified preparation of this product must be disposed after autoclaving or incineration after completion of task.
Keep container tightly closed. Stored in cool (10-30 ‘C) & Dry place.
Methyl Red and Voges-Proskauer test are among the two various tests used in the biochemical identification of bacterial species. These tests were originally studied by Voges, Proskauer and subsequently by Clark and Lubs to differentiate between members of the coli- aerogens group. Both the tests are based on the detection of specific breakdown products of carbohydrate metabolism.
All members of Enterobacteriaceae are, by definition, glucose fermenters. In MR-VP Broth, after 18-24 hours of incubation, fermentation produces acidic metabolic byproducts. Therefore initially all enterics will give a positive MR reaction if tested. However, after further incubation, required by the test procedure (2-5 days), MR – positive organisms continue to produce acids, resulting in a low pH (acidic) that overcomes the phosphate in buffering system and maintains an acidic environment in the medium (pH 4.2 or less).
