Principle:- Tryptone bile agar is another differential medium which specifically allows the Indole positive, late lactose fermenting E. coli. The Tyrptophan of Tryptone mediates the production of Indole which further can be detected by the application of the Kovacs or Ehrlichs reagent. While the other organisms get inhibited by the presence of Bile salts and incubating at elevated temperature of 44⁰C. The TBA is used for detection of E. coli by direct plating method over the Most Probable Number (MPN) Method.

Use:- Recommended for rapid detection and enumeration of Escherichia coli by modified direct platting method.

Directions:- Suspend 36.5 gram in 1000 ml distilled water adjust the pH if required. Boil to dissolve the medium completely. Tryptone Bile Agar sterilize by autoclaving at 15 lbs pressure (121 °C) for 15 min, cool it to 42-45 °C and distribute aseptically in to the plates. Ensure complete solidification and inoculate aseptically.

Specimens types analyzed:- Water samples, raw meat, milk and dairy, food samples, etc.

Performance and Evaluation:- The expected performance of the medium is liable to use as per the direction on the label when stored at optimum conditions and within expiry date.

Storage and Shelf Life:- Hygroscopic; Keep container tightly closed. Store at temperature below 30⁰C and dry place. Expected Performance when stored at optimum and within expiry date.

Disposal:- To avoid the contamination or propagation of any hazardous microbes the used, unusable or modified preparation of this product must be disposed after autoclaving or incineration after completion of task.

Precautions to be taken:- These microbial media are intended for the in-vitro use only. All the handling, experiments, storage, and discarding should be performed with the help of trained and experienced technicians and as per the established guidelines. The material should be disposed only after proper serialization by autoclaving.
