Principle :- Bushnell-Haas broth is described by Bushnell and Hass, for enrichment of hydrocarbon decomposing microorganisms. The media is composed of magnesium sulfate, calcium chloride, monopotassium phosphate, ammonium phosphate dibasic, potassium nitrate and ferric chloride. Magnesium sulfate, calcium chloride and ferric chloride provide necessary trace elements for growth of bacteria. Monopotassium phosphate and ammonium phosphate dibasic are buffering agents. Potassium nitrate is a nitrogen source. The media lacks carbon source and can be fortified with hydrocarbons for evaluation of hydrocarbon decomposing ability. Bushnell-Haas was recommended for the microbiological examination of fuels by the Society for Industrial Microbiology (SIM) Committee on Microbiological Deterioration of Fuels.

Use :- Recommended for isolation of hydrocarbon degrading microorganism and enumeration of fuels contaminating microorganisms.

Directions :- Dissolve 3.27 grams in 1000 ml distilled water.. Boil Bushnell Haas Broth to dissolve the medium completely and sterilize by autoclaving at 15 lbs. pressure (121°C) for 15 min, cool it to 42-45 °C and mix with sterile test hydrocarbon and distribute aseptically in desired and inoculate test sample aseptically.

Specimens’ types analyzed :- Hydrocarbons such as kerosene, light and heavy mineral oils, paraffin wax, and gasoline etc.

Precautions to be taken :- These microbial media are intended for the in-vitro use only. All the handling, experiments, storage, and
discarding should be performed with the help of skilled and knowledgeable technicians and as per the established guidelines. The material should be disposed only after proper sterilization by autoclaving. Please go through the MSDS of the media to avoid any accidents or in emergency.

Storage and Shelf Life :- Hygroscopic; keep container tightly closed. Store in cool dry place.

Disposal :- To avoid the contamination or propagation of any hazardous microbes the used, unusable or modified preparation of this product must be disposed after autoclaving after completion of task.
