Principle :- Clostridial agar is composed of tryptone, soya peptone, dextrose, sodium chloride, sodium thioglycolate, L-Cystine, sodium formaldehyde sulphoxylate, neomycin sulphate, sodium azide and agar. Tryptone and soya peptone provide nutrients and cofactors required for growth of clostridia. Dextrose is carbon source. L-Cystine is an amino acid enhances clostridial growth, Sodium chloride, sodium thioglycolate, L-cystine, sodium formaldehyde sulphoxylate maintain oxidation reduction potential of medium. Neomycin sulphate and sodium azide inhibits the growth of contaminating bacteria like Proteus, Pseudomonas and Bacillus. Agar is solidifying agent.
Use :- Recommended for the selective isolation of pathogenic Clostridia.
Directions :- Dissolve 47.00 grams in 1000 ml distilled water, boil Clostridial Agar to dissolve the medium completely and sterilize by autoclaving at 15 lbs. pressure (121°C) for 15 min, cool it to 42-45 °C and distribute aseptically in petri plates. Ensure complete solidification and inoculate test sample aseptically.
