PALCAM Agar Base – HX-MEDIA

Principle :- Listeria identification Agar base (PALCAM) is composed of peptone, starch, dextrose, mannitol, sodium chloride, ammonium ferric citrate, lithium chloride, esculin, phenol red and agar. Peptone provides nitrogen, vitamins and other essential nutrients. Starch, dextrose and Mannitol are source of carbon and energy source. Sodium chloride maintains osmatic equilibrium. Lithium chloride inhibits gram negative bacilli. Phenol red is pH indicator. The combination of mannitol and phenol red helps the detection of mannitol fermentation. Esculin is hydrolyzed by Listeria to form esculetin. This esculatin reacts with ammonium ferric citrate to form a brown-black complex seen as a black halo around colonies.

Use :- Recommended for isolation and identification of Listeria species from clinical and non-clinical samples.

Directions: Dissolve 34.44 grams in 500 ml distilled water. Boil PALCAM Agar Base to dissolve the medium completely and sterilize by autoclaving at 15 lbs. pressure (121 °C) for 15 min, cool it to 42-45 °C, aseptically add rehydrated content of 1 vial of Listeria Selective Supplement (RDM-LSS-01) mix well and distribute in sterile test tubes and inoculate test sample aseptically.

Specimens types analyzed :- Pharmaceutical samples, clinical and non-clinical samples etc.

Precautions to be taken :- These microbial media are intended for the in-vitro use only. All the handling, experiments, storage, and discarding should be performed with the help of skilled and knowledgeable technicians and as per the established guidelines. The material should be disposed only after proper sterilization by autoclaving. Please go through the MSDS of the media to avoid any accidents or in emergency.
