Principle :- Phenylalanine agar is composed of yeast extract, dipotassium phosphate, sodium chloride, DL-phenylalanine and agar. Yeast extract provides nitrogen, carbon, vitamins and other necessary growth factors. Dipotassium is buffering agent. Sodium chloride maintain osmatic balance. DL-phenylalanine is substrate for Phenylalanine deaminase, deaminate DL-phenylalanine to phenylpyruvvic acid. The synthesized phenylpyruvic acid is detected by addition of ferric chloride reagent. The ferric chloride reacts with phenylpyruvic acid to form green color complex. Agar is solidifying agent.
Use :- Recommended for differentiating phenylalanine deaminase producing Proteus and Providencia species form Enterobacteriaceae.
Directions :- Dissolve 23.00 grams in 1000 ml distilled water, boil phenylalanine agar to dissolve the medium completely and distribute aseptically in test tubes. Sterilize by autoclaving at 15 lbs. pressure (121 °C) for 15 min, cool it to 42-45 °C and allow the medium to solidify in a slanting position. Ensure complete solidification and inoculate test sample aseptically.
