De-Man Rogosa Sharpe Broth – HX-MEDIA – Hexa Biotech
De-Man Rogosa Sharpe Broth is described by DeMan, Rogosa and Sharpe (1960).
Principle:- Media composed of proteose peptone, meat extract (equivalent to beef extract), yeast extract, dextrose, sorbitan monooleate, ammonium citrate, sodium acetate magnesium sulfate, manganese sulfate and potassium phosphate dibasic. Proteose peptone, meat extract and yeast extract provide nitrogen, carbon, vitamins and other necessary elements.
Dextrose is carbohydrate source, Sorbitan monooleate, ammonium citrate, sodium acetate, magnesium sulfate and manganese sulfate are selective agents inhibits growth of other bacteria and essential for the growth of lactobacilli.
Directions:- Dissolve 55.15 grams in 1000 ml distilled water. Boil MRSB to dissolve the medium completely and sterilize by autoclaving at 15 lbs. pressure (121°C) for 15 min, cool it to 42-45 °C and inoculate test sample aseptically. Incubate in 5% CO2 atmosphere at 33-37°C for 24-72 hrs.
Use:- Recommended for cultivation of Lactobacillus species.
